膠原纖維(Collagen Fiber)是結締組織中分布最廣含量最多的一種纖維，廣泛分布于各種臟器，其中Ⅰ型膠原纖維主要是骨、皮膚、肌腱；Ⅱ型膠原纖維主要是軟骨膠原；Ⅲ型膠原纖維主要在胚胎組織、成人血管、胃腸道；Ⅳ型膠原纖維主要在基膜中。天狼星紅與其襯染液都是強酸性染料，易與膠原分子中的堿性基團結合，吸附牢固。偏振光鏡檢查，膠原纖維有正的單軸雙折射光的屬性，與天狼星紅復合染色液結合后，可增強雙折射，提高分辨率，從而區分兩型膠原纖維。

       改良天狼星紅染色液（膠原纖維染色），為G1472-試劑B，主要用于各種組織病變時對膠原纖維異常變化的研究。在普通光學顯微鏡下心臟血管等組織的膠原纖維被染成紅色，在偏振光鏡下對各種纖維化病變的分型和分級研究有一定的幫助作用。
